The validation data indicating the regulation of miRNAs on potential target genes can be obatined by detecting the luciferase activity.
Examples
The pmiR-RB-ReportTM h-AKT1 reporter gene plasmid vector (1 ug) and hsa-miR-146a mimic (50 nM) were co-transfected into A549 cell line, and the results of the dual luciferase assay kit showed that hsa-miR-146a pair AKT1 3’UTR has a significant regulatory effect.
The pmiR-RB-ReportTM h-BCL2 reporter plasmid vector (1ug) and hsa-miR-9 mimic (50nM) were co-transformed into A549 cell line, and the results of dual luciferase assay showed that hsa-miR-9 pair BCL2 3’UTR has a more pronounced regulatory effect.
